Ligation:
After 2-fold overdigestion with enzyme 75% of the DNA fragments can be ligated and 95% may be recut.

Non-specific hydrolysis:
Overnight digest with Bmr I is not recommended. A 50 μl reaction containing 1μg of DNA and 0.5 units of enzyme incubated for 4 hours resulted in the same pattern of DNA bands as a reaction incubated for 1 hour.
